Land-Ocean connections: Scientists discover how tree trunks, leaves and kukui nuts are indirectly feeding bottom fish in the submarine canyons off Moloka?i, Hawaii

ScienceDaily (Feb. 27, 2012) ? Scientists from the School of Ocean and Earth Science and Technology (SOEST) at the University of Hawaii -- Manoa (UHM) and colleagues recently discovered that land-based plant material and coastal macroalgae indirectly support the increased abundances of bottom fish in submarine canyons, like those off the north shore of Moloka'i. Less than a few miles from the shore, these underwater canyons connect to deep river valleys that cut across the landscape of north Moloka'i. The high elevation and forested landscapes along Moloka'i's north shore provide plant material, including decomposing tree trunks, leaves and tons of kukui nuts, which enter the ocean via river valleys and 'pile up' on submarine canyon floors.

"In areas surrounded by very low productivity waters, this steady supply of organic matter to the canyons supports relatively high abundances of macro-invertebrates that live associated with the seafloor (either on top or in the top layers within the sediments), such as polychaete worms (related to earth worms), tiny crustaceans (amphipods, tanaids and isopods) and mollusks (like tiny clam- and snail-like creatures). Those invertebrates in turn serve as food for many fish species that live and feed near the seafloor," says Fabio De Leo, lead author and PhD candidate at the UHM Department of Oceanography. "Areas surveyed outside the canyons showed very little contribution of plant and macroalgae material, providing support for our hypothesis that this material is really being channeled and accumulates mostly in the steep topography of the submarine canyons (i.e., a 'canyon effect')," completes De Leo.

De Leo, his PhD advisor (Dr. Craig Smith), colleagues from SOEST (Dr. Jeff Drazen), from the Hawaii Pacific University (Dr. Eric Vetter), and from the New Zealand National Institute of Water and Atmospheric Research (Dr. Ashley Rowden) used manned submersibles operated by the Hawaii Undersea Research Laboratory to perform numerous video transects in two submarine canyons off Moloka'i at depths ranging from 350 to 1,050 m (~1,000 to ~3,000 ft). Equipped with high-definition cameras and powerful light strobes the submersibles flew above the seafloor to quantitatively assess the presence of plant and macroalgae material and to count and identify all fish species.

This was among the first studies to quantitatively survey fish assemblages deeper than ~350 m around Hawaii, gathering more than 13 hours of video altogether. Among the most abundant species found were the so-called 'rattail' fishes (family Macrouridae) and the 'eel-like' fishes (synaphobranchids and halosaurs). Giant shark specimens were also observed but only when the submersible was stationary and not during the video transects. Therefore, that species was not included in the quantitative analysis. De Leo narrates how scary it was when he got 'face-to-face' with a 10-foot 'sixgill shark' with only the submersible's porthole separating his from the shark's nose, as the giant approached the submersible and hit the thick glass window with its face. "Wow, it was definitely a nerve-wracking sensation. I even dropped my video camera while trying to make good footage of the beast."

The researchers also report some unexpected results. They found that this 'canyon effect' of enhancing fish abundances is obliterated at intermediate depths (around 650 m) coinciding exactly with the core of an oxygen minimum zone (OMZ) that flows around the Hawaiian archipelago around those depths. An OMZ is a layer of seawater usually at subsurface that has depleted oxygen concentrations due to several biological and chemical processes, including excess respiration by microbes. This may be indicating that the bottom fish species are somewhat sensitive to the low oxygen concentrations in the water. This result corroborates previous studies that found low abundances and diversity of deep-sea scavenger animals at similar depths around Hawaii. "The expansion of OMZs in different parts of the oceans is of great concern among oceanographers, and it is thought to be one of the main effects related to climate/ocean warming." De Leo attended the annual Ocean Sciences Meeting in Salt Lake City last week and reports that the expansion of these low oxygen ocean layers was one of the hot topics discussed at the meeting. "We should definitely keep track of near future and long term effects of expansion of low oxygen zones and how this will affect a variety of marine organisms, including fish."

In hopes of understanding what other factors lead to an increase in the number and diversity of marine animals inside these abrupt topographic features, De Leo and colleagues are analyzing data on the abundance and diversity of macro-invertebrates that live associated with the seafloor in six other submarine canyons around Hawaii. There is an increasing body of evidence that suggests that canyons are a special type of topographic feature in the seafloor no matter in what oceanographic background they are located (i.e., highly productive continental margins or in a low productivity setting, such as an island margin like Hawaii). "With the goal of protecting ecologically important habitat and preserving biodiversity, this information is crucial when performing marine spatial planning and designing networks of Marine Protected Areas," says De Leo.

Study in Nature Medicine could lead to novel therapy for cancer

MAYWOOD, Ill. In a study published in Nature Medicine, Loyola researchers report on a promising new technique that potentially could turn immune system killer T cells into more effective weapons against infections and possibly cancer.

The technique involves delivering DNA into the immune system's instructor cells. The DNA directs these cells to overproduce a specific protein that jumpstarts important killer T cells. These killer cells are typically repressed in patients who have HIV or cancer, said Jos A. Guevara-Patino, MD, PhD, senior author of the study. Guevara is an Associate Professor in the Oncology Institute of Loyola University Chicago Stritch School of Medicine.

Guevara and colleagues reported their technique proved effective in jumpstarting defective immune systems in immuno-compromised mice and in human killer T cells taken from people with HIV.

Guevara said a clinical trial in cancer patients could begin in about three years.

The study involved killer cells, known as CD8 T cells, and their instructor cells, known as antigen-presenting cells. The instructor cells instruct CD8 T cells to become killer T cells to kill infected cells or cancer cells -- and to remain vigilant if they reencounter pathogens or if the cancer comes back.

In addition to getting instructions from the antigen-presenting cells, CD8 T cells need assistance from helper T cells to become effective killers. Without this assistance, the killer T cells can't do their job.

In patients who have HIV, the virus destroys helper T cells. In cancer patients, helper T cells also are affected. Among a tumor's insidious properties is its ability to prevent killer T cells from attacking tumors. It does this by putting helper T cells into a suppressed stage, limiting their ability to assist CD8 T cells, said Andrew Zloza, MD, PhD, one of the leading authors of the study.

In the study, snippets of DNA were delivered into skin instructor cells by a device known as a gene gun. The DNA directed the instructor cells to produce specific proteins, which act like molecular keys. When CD8 T cells interact with the instructor cells, the keys unlock the CD8 T cells' killer properties -- jumpstarting them to go out and kill pathogens and cancer cells.

With the use of this technique, the killer T cells would not need the assistance of helper T cells. So even if a tumor were to put the helper T cells in a suppressive cage, the killer T cells would still be able to go out and kill cancer cells. Researchers expect that future studies using the technique will make it applicable to many diseases, including cancer.

PHILADELPHIA Dual inhibition of vascular endothelial growth factor and c-MET signaling inhibited tumor invasion and metastasis in a laboratory model of pancreatic neuroendocrine cancer, according to a paper published in Cancer Discovery, the newest journal of the American Association for Cancer Research.

"Inhibition of VEGF signaling plus c-MET signaling results in a synergistic effect on tumors that leads to slowing of tumor growth and decreased invasiveness and metastasis," said lead researcher Donald M. McDonald, M.D., Ph.D., a professor at the University of California San Francisco Comprehensive Cancer Center.

Previous laboratory research had shown that inhibition of VEGF signaling with agents like bevacizumab or sunitinib can give rise to a number of side effects including increased tumor invasion and metastasis.

What was not known was whether anti-VEGF therapy results in elevated c-MET expression, which has been previously shown to promote tumor cell invasiveness and metastasis. To determine this, McDonald and colleagues conducted a two-phase laboratory study. They treated mice engineered to develop pancreatic neuroendocrine tumors with an anti-VEGF antibody, which reduced tumor size but increased invasiveness and metastasis. This treatment also increased tumor hypoxia and expression and activity of c-MET.

However, when both VEGF and c-MET signaling were inhibited simultaneously, there was a reduction in invasion and metastasis. The researchers tested three c-MET inhibitors: crizotinib and PF-04217903, which target c-MET but not VEGF signaling, and cabozantinib, a dual inhibitor that blocks VEGF and c-MET signaling.

McDonald said they conducted their initial study in neuroendocrine pancreatic tumors because the genetic mouse model of these tumors has been studied so extensively, and then observed the effect in other tumors as well.

"The intent of this study was to explore a mechanism, and there is no indication that this effect will be confined to pancreatic tumors," he said.

World on brink of carbon trade war over flights?

Countries opposed to a European Union law forcing the world's airlines to pay for greenhouse gas emissions have agreed on a basket of retaliatory measures, adding to a series of threats that have raised the prospect of the globe's first carbon trade war.

But the EU dismissed the threats as "hypothetical" and Russia's deputy transport minister also said the countries ? which include China, India and the United States ? were free to choose which of them they would actually carry out.

In Washington, the State Department said it would be "premature" to discuss specific retaliatory measures and voiced hope that the EU would reconsider its carbon emissions scheme.

The array of potential steps include barring national airlines from participating in the EU program, lodging a formal complaint with the United Nations' International Civil Aviation Organization (ICAO), ceasing talks with European carriers on new routes and imposing retaliatory levies on EU airlines.

"Every state will choose the most effective and reliable measures that will help to cancel or postpone the implementation of the EU ETS (Emissions Trading System)," Valery Okulov, whose ministry hosted the meeting attended by more than 20 nations, told a news conference.

Russia has a particular measure it can use against Europe. Okulov, the former chief executive of Aeroflot, said Russia could take out an old weapon against European carriers: overflight fees on routes over Siberia.

Forced to buy permits
Russia rescinded the onerous fees during the last decade, a move it linked to long-running talks for World Trade Organization membership, which it received late last year.

Since the start of this year, all airlines using EU airports are required to buy permits under the ETS, although they will not actually face a bill until next year. In addition, they will at first be handed 85 percent of allowances for free.

The European Commission reiterated it was standing by its law and said the Moscow meeting had delivered only negatives.

"Unfortunately, our question for Moscow meeting participants remains unanswered: What's your concrete, constructive alternative?" Climate Commissioner Connie Hedegaard said in a tweet.

Those opposing the program have debated the issue within the official ICAO framework and also in informal talks ? dubbed "the coalition of the unwilling" ? such as the two-day Moscow meeting, which ended on Wednesday.

Tensions grow over EU aviation emissions

Okulov said Saudi Arabia would organize the next unofficial meeting later this year.

Many nations and the EU have said the best arena to resolve the dispute would be the ICAO, which has been working on developing its own plan to curb rising aviation emissions.

U.S. tells EU: Stop
Mark Toner, a spokesman for the State Department, said Washington still hopes the EU will reconsider.

"The U.S. position on the inclusion of its airlines in this emissions trading scheme has been very clear and consistent. We believe that the EU needs to cease application of this scheme to foreign airlines and engage meaningfully with the International Civil Aviation Organization to find and develop a global approach to this problem," he said.

European court defies US over airline carbon tax

The EU has always said it would have no need to make all airlines using its airports pay for carbon under its ETS if a comprehensive global program were in place.

At a regular briefing in Brussels, Isaac Valero-Ladron, EU spokesman for climate action, reiterated that the EU "will review its legislation if there is an ambitious global agreement in force because we would be covered by this agreement."

Russia is among those to agree that the best forum would be the ICAO, but it still took a tough line at the Moscow talks.

"We have demonstrated our determination to ask for a cancellation or postponement of the EU ETS" in regard to airlines, Okulov said.

For all the talk, there were hints of a softening.

A list of signatories seen by Reuters showed only 23 nations, compared with 26 at earlier "coalition of the unwilling" talks.

'Don't want a trade war'
The final "Moscow joint declaration" also dropped proposals from an earlier draft, including sending a letter to the EU stating individual nations' inability to join the EU ETS, as well as a clause on reopening trade agreements in other sectors to pressure EU industries.

As had been expected by many observers, the meeting also stopped short of invoking the ICAO'S formal dispute resolution procedure, a lengthy process which could delay the quest for an ICAO-based global program.

But feelings are still running high over what some nations view as an infringement on sovereignty.

China's central government State Council, or Cabinet, earlier this month said all airlines were barred from taking part ? unless they had received government approval to do so.

The Commission has noted Chinese airlines have already signed up to a registry for eligibility for free allowances, while airlines have already begun passing on the extra costs to passengers through fare increases.

Representing the airlines, the International Air Transport Association called on the EU to be "sincere facilitators at ICAO" in the quest for a global framework.

"We don't want a trade war. But Europe's unilateral and extra-territorial EU ETS plans are clearly not acceptable to non-EU governments," Tony Tyler, IATA's director general and CEO said in remarks emailed to Reuters.

EU suspends copyright treaty ratification

(AP) ? The European Commission, facing opposition in city streets, on the Internet and in the halls of parliament, has suspended efforts to ratify a new international anti-counterfeiting agreement, and instead will refer it to Europe's highest court to see whether it violates any fundamental EU rights.

EU Trade Commissioner Karel De Gucht asserted Wednesday that an opinion from the European Court of Justice would clear away the fog of misinformation surrounding the Anti-Counterfeiting Trade Agreement, known as ACTA.

"This debate must be based upon facts and not upon the misinformation or rumor that has dominated social media sites and blogs in recent weeks," De Gucht told reporters in Brussels.

The decision appeared to reflect recognition by European Union officials of the political obstacles. Protests against the agreement were staged earlier this month in several European capitals ? including Berlin, Helsinki, Paris and Vienna ? by critics who say the agreement would stifle free speech and access to information.

The hacking group known as Anonymous claimed responsibility last week for a new series of hacks against the U.S. Federal Trade Commission and consumer rights websites. The sites were replaced with a violent German-language video satirizing ACTA.

ACTA has been under negotiation for years. Its drafters say it is needed to harmonize international standards to protect the rights of those who produce music, movies, pharmaceuticals, fashion goods, and a range of other products that often fall victim to piracy and intellectual property theft.

The U.S. has signed the agreement. Others include Australia, Canada, Japan, Morocco, New Zealand, Singapore, and South Korea. Mexico and Switzerland participated in the negotiations but have not yet signed.

The EU and 22 EU Member States signed ACTA on 26 January 2012 in Tokyo. Although the European Council ? the European Union heads of government ? unanimously approved ACTA in December, for the EU to be a party to the treaty, all 27 member countries would have to formally ratify it.

The treaty has been questioned by members of the European Parliament. On Sunday, European Parliament President Martin Schulz told Germany's ARD television: "I don't find it good in its current form."

He said the necessary balance between copyright protection and the individual rights of Internet users "is only very inadequately anchored in this agreement."

And in a statement Wednesday, David Martin, a spokesman on the issue for the Socialists and Democrats, the second largest bloc in the Parliament, welcomed the decision to get the court's opinion.

"Trade Commissioner Karel de Gucht today admitted that there are still many question marks about ACTA and what the implementation of the agreement, as it stands, would mean for citizens and for the freedom of the internet," Martin said. "The Parliament has been calling for more clarity for a long time, and we already requested legal opinions from several committees in the European Parliament. Now this ruling will be a good guarantee for the impact on fundamental rights.

EU officials say the agreement will not change EU law. They insist what was legal pre-treaty would remain legal the day after, and what was illegal would remain illegal. But they have said the EU must ratify it as an example to other countries where intellectual property rights are less protected than they are in the EU in order to protect European products and ideas from being stolen elsewhere.

"ACTA will not censor websites or shut them down; ACTA will not hinder freedom of the internet or freedom of speech," De Gucht said.

However, opponents fear ACTA would lead to censorship and a loss of privacy on the Internet. And the plan to gain court approval of the agreement has left at least some of them unimpressed.

"No legal debate can fix ACTA or give it a legitimacy that by design it cannot have," said Jeremie Zimmermann, co-founder of the Internet advocacy group La Quadrature du Net.

Coupe version of the updated performance GT car is revealed before its unveiling at the Geneva Motor Show.

Maserati is replacing its aging GranTurismo S with a new GranTurismo Sport, which will debut next month at the 2012 Geneva Motor Show.

The GranTurismo coupe gets some updated styling details from the outgoing S model. (Photo: Maserati) The car is essentially the coupe version of the equally stunning GranTurismo Convertible Sport launched last year and packs the same 460-horsepower 4.7-liter V8.

Visually setting the car apart from the current GranTurismo lineup are new, aerodynamically enhanced bumpers, revised headlights with LED detailing and darkened tail-lamp elements. The cabin also receives new seats and a sports steering wheel.

Power is sent to the rear wheels via a ZF six-speed automatic transmission or an optional sequential manual unit with the same number of gears. Both units also come with steering-wheel-mounted paddle shifters, though they are an option on the automatic.

In the lighter coupe body of the GranTurismo Sport, either drivetrain promises 0-60 mph acceleration in less than 5 seconds and a top speed in excess of 177 mph. Final numbers will be confirmed closer to the car?s Geneva debut.

Five days before her untimely death (cause as yet unknown, but a lethal combination of drugs and alcohol is suspected), Whitney Houston was in the recording studio working on new music. The song, ?Celebrate,? was written by R. Kelly and slated to play over the end credits to "Sparkle," Sony Pictures? forthcoming feature about a girl group formed by three teenage sisters, one played by American Idol winner Jordin Sparks, who duets with Houston on the track (though they did not record it together).

On Tuesday, Feb. 7, she arrived at producer Harvey Mason Jr.?s North Hollywood studio at around 5:30 p.m. ready to work. ?She's doesn?t lounge around,? Mason tells The Hollywood Reporter, still speaking in the present tense some 72 hours after Houston?s body was removed from her hotel room at the Beverly Hilton. ?The last session we did on Tuesday night, after we finished, we were dancing around the booth and laughing and just having a good time listening to what we had accomplished. Her demeanor the last two weeks was very upbeat. She was in a good place.?

Houston recorded two songs for "Sparkle," the other a gospel standard called ?His Eye is On The Sparrow,? and her voice, observed by many as shaky in recent years, was holding up for the most part during Tuesday?s four-hour session. ?Certain days are better than others,? says Mason. ?On Tuesday it was pretty good. We had been working on that song for a couple days. It took extra work because it's a duet. We weren't so much worried about the singing. We were spending more time on making sure the flow was right between the two artists and that each had their moment in the song.?

Indeed, Mason, who had worked with Houston since 1998, says that unlike most superstar artists, especially the so-called ?divas,? Houston was happy to share the spotlight. ?She was really adamant about making sure Jordin had enough time to shine,? he says. ?She kept saying, 'Let Jordin shine, that's her moment.' I thought that was pretty cool. She loves the new crop of singers. She's not the kind of girl who's competitive and mean to other singers.?

?But if another attempt at a musical comeback was in the works, you could say Houston was looking at another uphill battle. While 2009?s "I Look To You" sold well, with almost 1 million copies purchased in the U.S., landing at No. 29 on the year-end Billboard 200 chart, Houston?s inconsistent live performances made it difficult to stage a proper promotional blitz and a hit single did not emerge from the 11-song collection.

Still, Mason defends the album?s performance. ?Anytime you can go platinum and sell a million copies, you've accomplished something spectacular,? he says. ?I have a plaque on my wall from that record that I'm very proud of. I can't compare it to "The Bodyguard's" twenty million or some of those older records. It's such a different time. But if you're asking me, did it accomplish everything we set out to do? I would say that I think it's a great record. I'm not in control of what happens to the music once we turn it in to the label. Creatively, we did a great job. She did amazing work on the record. She was proud of it, and so was I.?

As 2012 rolled around, Houston was amped about new projects. She was keen on nabbing a seat on the "X Factor" judges? panel, for one. According to a show insider, Houston had reached out to "X Factor" brass that same week expressing her interest in the position. A meeting with Simon Cowell was set up for Monday, Feb. 13, two days after she died.

She was also excited about "Sparkle," the movie in which she acted, sang and was an executive producer. There was also talk of a sequel to "Waiting to Exhale" and a new solo album. ?She wanted to talk to Clive [Davis] about that,? says Mason, referring to Sony Music?s chief creative officer and Houston?s lifelong mentor. ?That's something I recognize coming away from this production -- she really had a lot coming up and a lot of things that she was looking forward to.?

Their time in the studio last week, says Mason, was just like any other of the many sessions he had done with the diva. ?She comes in, warms up, does a couple vocal exercises, usually says a prayer, listens to the song, asks me what I need and then she goes in the booth and we just work,? says the producer, who has also worked with the likes of Mary J. Blige and Beyonce. ?She?s always a sweetheart to me,? he says, shifting into present tense again. ?We get along great. We sometimes argue in the studio, but we always have a good time. She's fun-loving and we laughed a lot. We listened to a lot of music. She's such an amazing singer and always has been one of the best singers we'll ever know. My whole objective is to make her shine.?

Mason heard the news while he was at home having dinner with his daughter Saturday night. That?s when he received a call from A&R executive Larry Jackson, who had worked with Houston and Mason extensively over the years. ?I spent Saturday messed up over it,? Mason recalls, still affected. ?Sunday I was so bummed, I didn't really want to talk about it. Then I was taking my kid somewhere and one of the radio stations was doing a tribute. They played Whitney Houston songs all day. They weren't talking about anything, not her history or how she passed, they were playing her music. So I listened to her music for probably eight hours on Sunday. All the songs, all the vocals, the feeling you get when you hear them, all the memories they bring, the experiences you've had in your life while listening to those songs? That's what her legacy is going to be. No matter what anyone says, it will be the lasting memory of her for me and for probably most everybody.?

Mason?s last image of Houston was of the superstar singer dancing in the studio, looking happy and full of life. It?s an appropriate farewell for a singer who, he says, ?cared so much about making great music? and loved her fans. ??I remember she was always, like, 'I've got to do something better. I can't disappoint the people who buy my music.' Whitney had so much passion and energy and love for what she did? The best. The best.?
